Abstract

The design of efficient expression vectors is a major component in the development of a high level production system for recombinant antibodies. The BKV enhancer/Adenovims major-late promoter (BKV/AdMLP) and human cytomegalovims immediate-early enhancer/promoter (CMV) were evaluated. The Adenovirus tripartite leader containing a chimeric intron (AdTpl-SS), immunoglobulin leader intron and J/C chimeric intron were aided and/or shifted. Cell lines COS-1, CV-1, 293S, CHO and NSO were used as hosts for transient assays. The secreted products were quantified by ELISA.
