Abstract

We here report on 15N NMR investigations of ribonucleoside-3′-phosphates, biologically enriched with the 15N isotope at all nitrogen positions. The studies are carried out in water over a large pH range up to alkaline pH values resulting in a complete pH dependence of the 15N chemical shifts. With the 15N-enriched compound we are able to study the pH dependence of 15N-coupling constants. Together with the various 13C-15N couplings which are currently being investigated in our laboratory, these couplings should allow certain insights into the electronic structure of the nucleobases at different pH values. We also demonstrate that 15N NMR spectroscopy of the 15N amino groups is a good technique for the study of the exchange of amino protons with the protons of the solvent water. The pH dependence of the structure of the 15N amino resonance signal reflects the change of the exchange rate of the amino protons in the course of titration. In addition the interactions between pancreatic ribonuclease A and 15N-labeled pyrimidine-3′-phosphates are investigated using 15N NMR spectroscopy.
