Abstract

The last decade has seen a great research interest in the description and elucidation of the role of presynaptic receptors in neurotransmission, at both central and peripheral synapses. It is now evident that a large number of different receptor types may be present at presynaptic locations on the same nerve terminal and that activation of such presynaptic receptors may either facilitate or inhibit the release of transmitter from the neuron. For example, at peripheral adrenergic nerve terminals there is good evidence for inhibitory presynaptic receptors for α-adrenoceptor agonists, opiates, prostaglandins and muscarine agonists while facilitatory presynaptic receptors are present for β-adrenoceptor agonists and angiotensin II (Starke, 1977; Westfall, 1977; Langer, 1979; Muscholl, 1979; Stjärne, 1979). During the same period increasing evidence has emerged that adenosine and the adenine nucleotides (AMP, ADP and ATP) can also modify neurotransmission at a number of synapses by activation of presynaptic receptors.
