Abstract

Whenever new methods are developed for the quantitative analysis of amino acids the standard for comparison is nearly always the classic ion-exchange chromatography method developed in the 1950s by such workers as Stanford Moore, William H. Stein, D. Spackman, and Paul Hamilton [1,2,3]. When new analytical methods are compared to the classical ion-exchange method, too often claims of higher sensitivity and/or faster analysis times are made by comparison with ion-exchange systems 15–20 years old. Improvements in ion-exchange resins, use of small bore columns and the development of high-pressure injection valves and pumps have enabled researchers using the classic ion-exchange method of amino acid analysis to decrease the analysis time from 24 hours in 1958 to less than 30 minutes, and to improve sensitivity by more than six orders of magnitude. Currently, ion-exchange chromatography of amino acids is just one of many techniques available in the rapidly expanding field of high performance liquid chromatography (HPLC).
