Abstract

Collections of seed and clonal material are traditional ways of storing genetic resources, while tissue cultures provide a complementary way. Freeze-preservation of cultured cells and meristems has become popular for long-term germplasm preservation (7,28,53). However, freeze-preservation requires the use of sophisticated techniques and expensive equipment, and often leads to tissue damage during freezing and thawing cycles. Another way of storing germplasm which does not require transfers is to limit growth by modification of the medium, e.g., by adding osmotically active substances. Cold storage of cultures at 0 to 15°C is a third method.
