Zusammenfassung
Als Ursache von Depressionen werden Serotonin- oder Katecholaminmangelzustände an spezifischen Rezeptoren im zentralen Nervensystem diskutiert [10]. Dieser Theorie entspricht der Wirkmodus der klassischen Antidepressiva, die die Wiederaufnahme von Serotonin und/oder Noradrenalin in die präsynaptische Nervenendigung hemmen oder den enzymatischen Abbau der Monoamine, beispielsweise durch eine Hemmung der Monoaminoxidase, unterdrücken. Beide Effekte führen akut zu einem erhöhten Angebot an Monoaminen. Durch die Veränderung der Monoaminkonzentration im synaptischen Spalt sowie durch Rezeptorblockade soll nach längerer Behandlungsdauer die Dichte der Neurotransmitterrezeptoren verändert werden. Allerdings reichen diese Mechanismen nicht aus, um die Wirksamkeit aller Antidepressiva zu erklären. Sie begründen auch nicht die Latenzzeit von mehreren Wochen, die zwischen dem Beginn der Pharmakotherapie und dem Eintreten der stimmungsaufhellenden Wirkung liegt [10].
