Fluorometric Measurement of Callose and Other 1,3-β-Glucans

Abstract

The term callose originates from cytochemical observations. Over the last hundred years many examples have been reported where staining and fluorescence techniques reveal that certain cell-wall appositions appear to consist of substances distinct from the materials which usually constitute the cell walls of higher plants (for a review on the early microscopical work see Eschrich 1956). Later it was shown that these staining properties are due to the presence of essentially linear l,3-β-glucans (for citations see Fincher and Stone 1981; Evans et al. 1984), although some other linkages may be present (Hinch and Clarke 1982). The term callose is now used synonymously for linear l,3-β-glucans exhibiting a degree of polymerization high enough to render the polymer water-insoluble.
