Studies of Acyclovir and Ara-AMP Against Herpetic Keratitis in Rabbits

Summary

The therapeutic effects of 3% adenine arabinoside 5′-monophosphate (ara-AMP), 3% acyclovir (ACV) and 0.5% 5-iodo-2′-deoxyuridine (IDU) ointment on herpetic keratitis and their toxic effects on normal cornea in rabbits were compared. Ara-AMP showed the strongest antiviral activity, but it produced corneal erosion after 4 days application. Acyclovir showed an equal therapeutic effect to that of IDU and was found to be non-toxic. Acyclovir was, therefore, concluded to be the most effective for treatment of herpetic keratitis.

Zusammenfassung

Im Kaninchenmodell untersuchten wir vergleichend den therapeutischen und den toxischen Effekt von 3% Adeninarabinosid Monophosphat (Ara-AMP), 3% Acyclovir (ACV) und 0,5% IDU bei der Herpeskeratitis und bei Normalaugen. Ara-AMP erwies sich als stärkstes Virustatikum, führte aber nach 4tägiger lokaler Anwendung zu Hornhauterosionen. Acyclovir war annähernd so wirksam wie IDU, aber viel weniger toxisch als letzteres. Deshalb schließen wir, daß Acyclovir von den getesteten Virustatika das beste zur Therapie einer Herpeskeratitis ist.
