Ultrastructural Observations on Isolated and Cultured Protoplasts of Lily Pollen

Abstract

Pollen protoplasts were obtained from pollen grains of Lilium longiflorum by dissolving intine layers 2 and 3 with treatment of macerozyme (pectinase) and cellulase. The protoplasts regenerate their cell walls by secretion of polysaccharides from vesicles derived from Golgi bodies. The membranes of these secretive vesicles are reformed in the cytoplasm by the membrane flow system. The ultrastructures of an isolated generative cell and its protoplast are shown.
