Acute Dose Response of Isosorbide-5-Mononitrate in Patients with Effort Angina

Abstract

Isosorbide-5-mononitrate (IS-5-MN) has a predictable pharmacokinetic profile; it is nearly 100% bioavailable after oral dosing, does not undergo first pass metabolism, has a plasma half-life between 4 and 6 h, and has dose-linear kinetics between 10 and 80 mg [1–3]. A predictable pharmacokinetic profile is an advantage when studying dose response and designing dosage regimens to avoid nitrate tolerance.
