Abstract
The use of plant protoplasts for genetic manipulations has been the central theme for most of the applications of protoplast technology. Significant progress has been made in various aspects of genetic manipulations using such protoplasts. This progress will be briefly surveyed. Increasingly, however, it is becoming realised that a better understanding of the physiological properties of protoplasts will greatly enhance our ability in this respect, and these aspects will be highlighted.
