Summary
The protective effect of O-(β-hydroxyethyl)-rutosides (HR) against the damaging action of etacrynic acid and polidocanol was studied with vascular endothelium (hind leg of anaesthesized cat), smooth muscle preparations of blood vessels (isolated portal vein, aorta of the rat), and with the ileumot the guinea-pig. These studies were intended to investigate the unspecificity of the protective action of HR and its oral efficacy. The following results were obtained:

HR protected not only the vascular endothelium but also smooth muscles of vasculature and ileum.

When HR is added to the isolated organs in vitro, it can be washed out easily. When given i. v. before isolation of organs, the protective activity cannot be washed out, even the organs are washed several times. Thus it is suggested that HR is bound to the tissue of the isolated organs.

This is in agreement with data showing very short elimination half-lives of HR in plasma, which are detected mainly after i. v. administration and prolonged half-lives of the fractions of HR stored in tissues. These fractions are probably responsable for the protective activity of the drug.

In our hands HR is active after i. v. and oral administration. Using a single i. v. administration considerably higher dosages are required compaired to repeated oral administration. Reasons for these results may be: (a) Very short elimination half-lives in plasma and prolonged half-lives in tissues, (b) an active transport of HR which facilitates absorption into the blood stream after oral administration.
Zusammenfassung
Der protektive Effekt von O-(β-Hydroxyethyl)-rutosiden (HR) gegenüber der schädigenden Wirkung von Etacrynsäure und Polidocanol wurde am Gefäßendothel (Versuche am Hinterlauf der narkotisierten Katze), an der glatten Gefäßmuskulatur (isolierte Pfortader und Aorta der Ratte) und an der glatten Darmmuskulatur (Ileum des Meerschweinchens) untersucht. Ziel der Untersuchungen war der Nachweis einer Unspezifität der protektiven Wirkung des HR und seiner Wirksamkeit bei oraler Applikation. Die Untersuchungen brachten folgende Ergebnisse:

HR wirkt protektiv nicht nur am Gefäßendothel, sondern ebenso an der glatten Muskulatur der Gefäße und des Darmes.

HR ist bei Versuchen an den isolierten Organen nur dann leicht auswaschbar, wenn es in das Organbad gegeben wird. Dagegen haftet es an den isolierten Organen fest, wenn es vor deren Entnahme i.v. in das Tier gegeben wurde. Die Wirkung bleibt dann trotz wiederholten Spülens unvermindert erhalten.

Dies steht im Einklang mit der sehr kurzen Plasmahalbwertzeit, die vor allem bei i. v. Applikation eine Rolle spielt, und der sehr langen Halbwertzeit des Anteils, der im Gewebe verankert ist und dort protektiv wirkt.

HR ist sowohl bei i. v. als auch bei peroraler Applikation wirksam. Bei i. v. Applikation erfordert die Wirkung im akuten Versuch erheblich höhere Dosen als bei peroraler. Als Ursache hierfür werden zwei Mechanismen diskutiert: zum einen die sehr kurze Plasmahalbwertzeit und die lange Gewebehalbwertzeit, zum anderen ein aktiver Transport, der möglicherweise bei peroraler Anwendung die Aufnahme in die Blutbahn fördert.
