Calcium-mediated Transduction of the Hormonal Message in 1-Methyladenine-induced Meiosis Reinitiation of Starfish Oocytes

Abstract

In most animals, oocytes develop to the prophase stage of meiosis and then stop. In starfish meiosis is reinitiated by a hormone produced by the follicle cells (Hirai et al., 1973), which has been identified as 1-methyladenine (1-MeAde) (Kanatani et al., 1969). Fifteen to 20 min after 1-MeAde addition to a fully grown prophase-blocked oocyte, nuclear membrane breaks down, then chromosomes condense and the meiotic spindle is formed. When cytoplasm taken from starfish oocytes treated for more than 7 min with 1-MeAde is injected into prophase-blocked oocytes, meiosis is reinitiated (Kishimoto and Kanatani, 1976). The maturation-promoting factor (MPF) present in cytoplasm is not depleted through multiple serial transfers, whereas it loses its activity when diluted in its extracted state: this suggests that it exhibits ‘autocatalytic properties’. MPF lacks specificity since for example amphibian MPF can induce meiosis when injected into starfish oocytes (Kishimoto, unpubl. data). On the other hand, cytoplasmic extracts of synchronized Hela cells can induce meiosis when injected into Amphibian oocytes. Extracts from G1 and S phase cells have no activity whereas MPF activity increases rapidly during late G2 to reach a peak in mitosis (Sunkara et al., 1979). These findings support the view that in the animal kingdom a common cytoplasmic factor might control both the release of oocytes from prophase block and the progression of mitotic cells from G2 to M. This paper is concerned with the analysis of some significant events occurring in starfish oocytes between interaction of 1-MeAde with the cell membrane and appearance of MPF.
