Abstract

Isolated plant protoplasts have become an important experimental system in some physiological, biochemical and genetic works. However, many experimental protoplast systems have proved to be limited by one or more technical problems, requiring extensive and sometimes fruitless investigation. This review focuses on technical problems and recent advances in isolation and cultivation of protoplasts. Earlier reviews have focused on different aspects of the handling of plant protoplasts. (Cocking, 1972; Gamborg et al., 1973; Eriksson et al., 1974). The first International Symposium on Higher Plant Protoplasts in Versailles, 1972, was convened to discuss significant advances and the basic difficulties in handling isolated protoplasts for cell transformation and cell fusion (Tempe, 1973). Vasil (1976) has in his review focused on progress, problems and prospects of plant protoplast research.
