Abstract
The theory that viruses cause insulin-dependent diabetes mellitus (IDDM) has been advanced sporadically for more than a century. During the past decade this theory has attracted renewed interest following the description by Craighead and McLane (1968) of a murine model of IDDM induced by the M variant of encephalomyocarditis (EMC) virus. An intriguing aspect of this model, which recapitulates many of the metabolic and pathologic features of human IDDM, is that EMC viruses are picomaviruses closely related to certain human pathogens, specifically coxsackieviruses. Although there is little evidence to suggest that EMC viruses per se cause human diabetes, efforts to incriminate Coxsackieviruses in IDDM have met with considerable success. In this chapter, we will review the evidence linking viruses to the etiology of IDDM.
