Summary

The effect of temperature on enzymatic synthesis of cephalexin (CEX) from 7-amino-3-deacetoxycephalosporanic acid (7-ADCA) and D-alpha-phenyl glycine methyl ester (PGM) using immobilized penicillin G amidohydrolase (E.C.3.5.1.11) from E. coli ATCC 9637 was investigated. It was found that decreasing temperature results in a marked enhancement in both yield in condensation product and stability of catalyst. Analogous results were obtained using 7-amino-3-chloro (deacetoxymethyl) cephalosporanic acid as substrate for enzymatic synthesis of cefaclor.
