Abstract
5-lipoxygenase (5-LO) is expressed in myeloid cells like granulocytes, monocytes macrophages and in B-lymphocytes. In addition, 5-LO expression was reported for neuronal tissues and keratinocytes. In myeloid cells, expression of the 5-LO pathway (5-LO) is upregulated during cell maturation. It has been shown by various authors that differentiation of HL-60 cells is accompanied by an upregulation of 5-LO mRNA, protein, and activity (Bennett et al., 1993; Kargman and Rouzer, 1989). Depending on the differentiation inducer, 3–6-fold differences in 5-LO mRNA levels were observed for HL-60 cells (Bennett et al., 1993; Steinhilber et al., 1993a). The combination of 1,25-dihydroxyvitamin D3 (calcitriol) and transforming growth factor-ß (TGFß), either alone or in combination with dimethylsulfoxide, was the most prominent inducer of the 5-LO pathway, leading to cellular protein expression and activity comparable to (or higher than) what is found for normal human granulocytes (Brungs et al., 1994; Steinhilber et al., 1993b).
