"Factitious Angiogenesis": Endothelialization of Artificial Cardiovascular Prostheses

Abstract

There is an increasing demand for permanent cardiovascular prostheses, such as vascular grafts, cardiac assist devices or total artificial hearts. Currently, their longterm usage is thwarted by a number of divergent problems, such as thromboembolism, or neointimal hyperplasia. These complications can be frequently traced to inadequate hemocompatibility of the synthetic, blood contacting surfaces. This problem maybe ameliorated by factitious angiogenesis, namely the generation of nonthrombogenic, artificial blood conduits by precoating their luminal surfaces with a functional monolayer of autologous endothelial cell. In this presentation we will discuss the endothelialization of three cardiovascular prostheses: small caliber vascular grafts, artificial blood pumps, and autologous skeletal muscle ventricles.
