Abstract

Intense clinical and experimental interest continues to be placed on cancer invasion and metastasis since these pathologic processes are widely considered to be the major cause of death resulting from malignant disease (1–3). In addition, the properties of invasion and metastasis define the malignant phenotype of tumor cells (1–3). Tumor cells with metastatic potential, which comprise only a subpopulation of the heterogenous cells within the primary neoplasm, can only give rise to secondary metastatic colonization following numerous and complex tumor cell-host interactions (4); for example, invasive, metastatic cells penetrate a number of host extracellular matrices and barriers. This chapter will review recent findings dealing with the interaction of malignant cells with one particular type of extracellular matrix that is encountered by invasive tumor cells at several steps of the metastatic process: the basement membrane. Following an overview dealing with the structure and function of basement membranes, including vascular basement membranes, emphasis will be placed on the discrete steps involved in tumor cell-basement membrane interaction: tumor cell attachment, basement membrane dissolution mediated by tumor cell-derived proteolytic enzymes and subsequent tumor cell locomotion through localized domains of degraded basement membrane in the proximity of invading tumor cells.
