Chronic Administration of Monoamine Oxidase Inhibitors: Basic and Clinical Investigations

Neuropsychopharmacology of the Trace Amines

Abstract

Tranylcypromine (Parnate) and phenelzine (Nardil) are frequently prescribed monoamine oxidase (MAO)-inhibiting antidepressants. Although there have been a large number of reports on the effects of these drugs on biogenic amines in laboratory animals, in many cases these studies concentrated on either the classical neurotransmitter amines or the trace amines and did not measure simultaneous changes occurring in both classes of amines. In addition, many of these reports dealt with effects on amines at short time periods after drug administration rather than considering the effects of long-term administration. In our laboratories we have been investigating the effects of long-term administration of these drugs on brain concentrations of several amines in rats and have carried out preliminary studies on the effects of 2-week administration of phenelzine on urine and plasma levels of some amines, their amino acid precursors and their metabolites in patients with dysthymic disorder. Some of these findings will be reported here.
