Summary
The reaction between adenovirus hemagglutinins (virion, dodecon and penton) and red blood cells (RBC) was investigated. Purified isotope-labelled virions, dodecons and pentons of human adenovirus type 3 (Ad 3) (subgroup I) or virions of Ad 2 (subgroup III) were mixed with monkey or rat RBC and the radioactivity associated with RBC under different conditions was measured. The following results were obtained:

The number of Ad 3 receptors on monkey RBC seemed to be larger than that of Ad 2 receptors on rat RBC.

At 37° C, Ad 3 virions and dodecons attached to monkey RBC more rapidly than Ad 2 virions to rat RBC. Almost no attachment of isolated Ad 3 monomer penton was detected but the addition of anti-vertex capsomere antibody prompted an attachment. At 4° C, the attachment of Ad 3 virions and dodecons was slower than that of Ad 2 virions.

RBC-associated Ad 3 virions and dodecons, but not Ad 2 virions, were released from RBC by reducing the temperature to 4° C. When the reaction mixture was warmed again to 37° C, the released dodecons re-attached quantitatively to RBC.

RBC-associated Ad 3 virions and dodecons and Ad 2 virions were removed from RBC by the addition of antibody against homologous hemagglutinins. Antibody against Ad 3 hexon also caused a detachment of Ad 3 virions, but not of Ad 3 dodecons.

Addition of large amounts of nonlabelled hemagglutinins of homologous and heterologous types belonging to the same subgroup caused a detachment of RBC-associated labelled virions and dodecons.
