Abstract

Next-generation sequencing (NGS) technologies refer to the high-throughput sequencing techniques. “Next-generation” sequencing technologies lead to rapid generation of data by sequencing massive amounts of DNA in parallel using diverse methodologies with definite sequencing platform, say Illumina or Agilent. The basic methodologies involve, RNA isolation, cDNA preparation followed by cDNA library preparation. NGS experiments basically consist of four phases: Sample collection, Template generation, Sequencing reactions and detection, and Data analysis. NGS experiments can be grouped broadly into two general categories: de novo assembly and resequencing. Evolution of NGS technology has led to advances in genomics far better than DNA microarray. In this book, we have discussed a number of technologies pertaining to NGS. The examples include epigenetics, metagenomics, exome sequencing, genome wide association studies, transcriptomics, and RNA-seq.
