Summary
Analysis of the Gc system was carried out by isoelectrofocusing on polyacrylamide gels. The six common phenotypes Gc IS, 1 F-lS, 1F, 2-1S, 2-1F, and 2 were observed, and, in addition, the rare variants Gc 1A 5, 1C3 and two new Gc 2 variants. In a sample of 1523 unrelated individuals from Southern Germany the following allele frequencies were found Gc*lS = 0.5975, Gc*lF = 0.1402, Gc*2 = 0.2610, Gc*Var = 0.0013. The assumed autosomal codominant mode of inheritance was confirmed by the examination of 349 pairs of parents with a total of 432 children. Analysis of 306 cases of disputed paternity revealed 52 exclusions of paternity in the Gc system. In two cases the exclusions were found in the Gc system only, while in the remaining cases further exclusions were obtained with blood groups and/or serum protein and enzyme systems. For paternity testing two different classification systems were employed. For classification of Gc subtypes isoelectrofocusing followed by immunofixation was used.
Zusammenfassung
Die Untersuchung des Gc-Systems erfolgte mittels der isoelektrischen Fokussierung auf Polyacrylamidgelen. Es wurden die sechs häufigen Phanotypen Gc 1 S, 1 F-l S, 1F, 2-1S, 2-1F und 2 sowie die seltenen Typen Gc 1A5, 1C3 und zwei neue Gc- 2-Varianten beobachtet. für eine Stichprobe von 1523 nicht verwandten Personen aus Siiddeutschland ergaben sich folgende Allelfrequenzen: Gc*l S = 0,5975, Gc*l F = 0,1402, Gc*2 = 0,2610, Gc*Var = 0,0013. Der angenommene autosomal kodominante Erbgang der Subtypenallele wurde durch die Untersuchung von 349 Elternpaaren mit insgesamt 432 Kindern bestatigt. Die Untersuchung von 306 strittegen Ein- und Mehrmannfällen ergab 52 Ausschlüsse im Gc-System. Mit Ausnah me von zwei isolierten Gc-Ausschliissen decken sich diese Ausschlüsse mit denen anderer Blutgruppen-, Serumprotein- und Enzymsysteme. für die Paternitatsbegutachtung wurden zwei unterschiedliche Bestimmungsmethoden verwendet. Zur sicheren Typisierung der Gc-Untergruppen wurde nach der Isoelektrofokussierung eine Immunofixation durchgefuhrt.
