Summary

A four-step purification system using high performance liquid chromatography (HPLC) was developed and used in conjunction with a rapid and sensitive bioassay to isolate and purify a number of myotropic peptides present in methanolic head extracts of the cockroach, Leucophaea maderae, and the house cricket, Aceta domesticus. The extracts were sequentially fractionated on a series of four HPLC columns exhibiting different separation characteristics. The procedure yielded peptides of sufficient purity for primary sequence determination. A total of 17 unique peptides, including the first gastrin/cholecystokinin-like invertebrate peptides, were structurally characterized and synthesized.
