Abstract

Obesity is a growing epidemic where an abnormal accumulation of fat leads to severe pathological conditions and it recently emerges as an avoidable risk factor for cancer. Obesity fine-tunes the activation of AMPK and PI3K/AKT pathway to induce cancer cell survival, proliferation, metabolism, and genome stability by modulating insulin growth factor signaling, leptin signaling, adiponectin signaling. Autophagy regulates diverse aspects of cellular metabolism and its impairment is associated with obesity-related pathophysiology and its subsequent progression to cancer. Autophagy reduces obesity-induced metabolism dysfunction, inflammation, lipotoxicity, oxidative stress, and proteotoxicity. Autophagy plays a puzzling role in cancer; it prevents cancer initiation in the early stages while leads to cancer development and persistence in later stages. This chapter would link pathways for autophagy dysregulation in obesity and its role in subsequent cancer progression. Accumulated pieces of evidence suggest that modulating autophagy could be an attractive therapeutic intervention for the treatment of obesity-induced cancer.
