Polar Lipids in Spinach Leaf Mitochondria

Abstract

Methods for preparation of highly purified mitochondria have made it possible to determine the lipid composition in mitochondria from spinach leaves. The chlorophyll contamination in the preparations was very low, yet some galactolipids were found (about 3 µg·mg−1 mitochondrial protein). The phospholipid composition of spinach leaf mitochondria was similar to the phospholipid composition reported for mitochondria from both photosynthetic and non-photosynthetic tissue. C18:2 and C18:3 were the dominating fatty acids in all phospholipids except for phosphatidylglycerol and phosphatidylinositol where high amount of C16:0 were found. The only significant difference between spinach leaf mitochondria and mitochondria from non-photosynthetic tissue were found to be the lower amount of total acyl lipids on a protein basis in the spinach mitochondria.
