Levels of genetic differentiation in Pinus halepensis Mill. in Spain using quantitative traits, isozymes, RAPDs and cp-microsatellites

Abstract

The pattern of geographic variation and the relationship among genetic variation in quantitative and molecular traits have been investigated in 15 populations of Aleppo pine covering the range of its distribution in Spain. The FST was estimated by means of 5 polymorphic isozyme loci, and, in a subset of 6 populations, by 24 RAPDs and 9 Cp-microsatellites loci. The phenotypic traits measured were the following: germination parameters obtained by the Gompertz function, height growth at age of 4 years in two provenance-progeny test and height growth phenology parameters in one of the two sites. The average FST value estimated using isozymes analysis (0.11) was compared with that obtained from phenotypic traits. Results showed a weak selection acting in some of the traits (phenology) and a strong selection in germination and height growth. Results obtained when analysing 15 populations were comparable with those obtained by RAPDs and Cp-microsatellites in a smaller subset of populations. The importance of selection in the pattern of variation of the species was discussed.
