Abstract
Direct methods for solving the phase problem in X-ray crystallography were not supposed to work for macromolecules, or on reflection intensity data which do not reach atomic resolution. As a consequence, young protein crystallographers may not have been exposed to direct methods as much as colleagues in the field of “small molecules”.
