Elucidation of metabolic network of mammalian cell line K562 using 2-14C glucose label and stoichiometric modelling

Abstract

Nutrient consumption and incorporation of the label from 2-14C glucose into metabolic byproducts were studied. A compartmentalised metabolic network of K562 cells was assembled and intracellular fluxes using a stoichiometric model were calculated. An equation system of label movement was created. Enrichments of lactate, alanine, glutamate and proline predicted from the model agreed well with those experimentally determined. According to the model 5% of glucose consumed entered the mitochondria. Most of the pyruvate in the mitochondria was derived from glutamine. Alanine was formed in mitochondria.
