Rapd-Scar-Sscp: A Method to Detect Molecular Differentiation in Closely Related Oak Species

Abstract

Quer eus petraea and Quer eus robur are two closely related interfertile oak species, that showed extremely low genetic differentiation in previous studies, although morphological and ecophysiological differences were reported. We develop a technique to detect genomic regions, evenly rarely dispersed in the genome, involved in molecular interspecific differences. The method is a three step procedure: (1°) random amplification with 10 mers to detect discriminant fragments (2) conversion of informative RAPD fragmenst into SCAR fragments and (3) detection of polymorphisms within SCAR fragments by using SSCP. The method (RAPD-SCAR-SSCP) is applied to the Q. petraea — Q. robur example with particular emphasis to the construction of SCARs and allelic interpretation of SSCP fragments. Genomic regions differentiating the two species are extremely rare (less than 1 % to the total number of fragments screened) and highly polymorphic (13 to 19 alleles for fragmenst varying in size from 450 to 790 bp).
