Studies on Synaptosomes: New Insights on Calcium Antagonists

Abstract

The use of cerebral synaptosomes permits investigation of certain aspects and consequences of calcium metabolism by the brain not readily accomplished in other biological samples. Since alterations in brain calcium homeostasis appear to play a critical role in cell damage following anoxic/ischemic insult (1,2) use of synaptosomes may provide an important investigative tool for the in vitro study of this aspect of brain anoxia and ischemia. Neuronal cell culture permits study of some mechanisms of calcium toxicity as well as the neurotoxic potential of excitatory amino acids (EAA) in ischemia, but does not readily permit study of antecedent presynaptic events. Microdialysis techniques are able to monitor EAA release following ischemia (3), but the poor time resolution of the measurements precludes analysis of the time course or kinetics of EAA release, nor can the cell of origin of the compounds be readily discerned.
