Abstract
Studies of drug binding to DNA have revealed much about the properties of the macromolecule, from effects on its supercoiling to kinetic factors involved in nucleotide sequence recognition and determinants of base pair discrimination. A spin- off has been the realisation that particular base pair substituents such as the 2-amino group of guanine strongly affect the disposition of ligand binding sites, the width of the helical minor groove, the flexibility of the polymer, its interaction with proteins, and the winding of DNA round the histone octamer in nucleosome core particles.
