Structural Studies on the Yeast tRNAPhe by the 1H NMR of Modified Bases. Effect of Removal of the Y Base and Anticodon Binding

Nuclear Magnetic Resonance Spectroscopy in Molecular Biology

Abstract

Recent studies of Kan et al. [1] and Robillard et al. [2] monitoring the methyl and methylene resonances of yeast tRNAPhe in the high field region of the spectra have demonstrated that these resonances are very sensitive to structural changes which occur upon heating or chemical modification. Furthermore the assignment of these resonances is less ambiguous than is the assignment of H-bonded resonances. In this communication we report the assignments of the modified bases in yeast tRNAPhe at low temperature, and the effect of removal of the Y base from the anticodon loop of yeast tRNAPhe on the resonances of minor nucleosides.
