Abstract

The use of bacterial genetic techniques to study the Rhizobium-soybean symbiosis has been hampered by the slow growth rate of Bradyrhizobium japonicum, the classical endosymbiont of soybeans, and by the apparent absence of symbiotic plasmids in this organism. The discovery of fast-growing R. japonicum strains from China (Keyser et al., 1982) raised the possibility that these strains may be better candidates for such genetic studies. In this paper, we summarize the results of our mutagenesis, plasmid mobilization, cloning, and DNA sequencing studies of sym plasmid genes that are required for nodulation and nitrogen fixation in fast-growing R. japonicum.
