Effect of 6 month treatment with a gonadotrophin releasing hormone agonist analogue on endocrine and histological features of human testis tissue

Abstract

The paradoxical antigonadotrophic effects of highly potent GnRH analogues (for a recent review, see Reference 1) have provided a new tool for inhibition of the pituitary-gonadal function. Besides certain diseases where inhibition of the pituitary-gonadal function is the goal of treatment (e.g. sex-hormone-dependent malignancies and precocious puberty), the same effect may be desirable in the development of new contraceptive methods in the male, in combination with treatment with testosterone (T) to prevent the loss of libido and impotence2,3. Apart from decreased circulating gonadotrophin and gonadal hormone levels, very little is known in the human about the gonadal effects of GnRH agonists in the long term. We found it of importance to assess the specific effects of long-term GnRH agonist treatment on morphology and hormonal function of human testis tissue. Such information may appear useful in assessment of the feasibility of GnRH agonists for the development of male contraception. The preliminary data of this study are reported here.
