Abstract

The successful application of high resolution NMR methodology to human metabolism studies, whether for research or clinical-diagnostic purposes, requires the development and refinement of methods for obtaining spectra from local regions within the subject. As yet no quantitatively perfect localization method has been found, although a several somewhat imperfect methods, such as surface coil and topical magnetic resonance techniques have been used, and a number of new methods based on proton imaging technologies have been suggested. This discussion critically reviews the surface coil and topical magnetic resonance methods which have been employed for high resolution spectroscopy of animal tissues. It goes on to stress that high resolution spectroscopy of small metabolites has special limitations which will be important in defining the effectiveness of those methods which have been suggested, but not yet applied in living animal experimentation.
