Structure of the Alzheimer Paired Helical Filament

Abstract

In senile dementia of the Alzheimer type three principal kinds of fibrillary deposit occur in the brains of affected individuals. These comprise neurofibrillary tangles, neuritic plaque cores and cerebrovascular amyloid. All three lesions consist of dense fibrous aggregates and have been called amyloid on the basis of the histopathological test of showing green birefringence when stained with Congo red dye (1). However, the principal fibrous component of the tangle, the paired helical filament (2), is ultrastructurally quite distinct from the fibrils found in cerebrovascular amyloid deposits or in plaque cores (3) or from the fibrils found in other types of amyloidosis (4). This paper will deal with the detailed structure of the paired helical filament and compare it with the amyloid-like fibrils of the plaque core.
