Zusammenfassung
Es wird eine in vivo Methode zur isolierten Leberperfusion bei der Ratte vorgestellt, die die Durchführung von Chemotherapiestudien an transplantablen und chemisch induzierten Lebertumoren ermöglicht. Nach Kanülierung der Arteria hepatica propria und der Vena cava inferior und vollständiger Isolation der Blutversorgung der Leber erfolgt eine Perfusion mit dem Nitroso-Harnstoffderivat Hydroxyethyl-CNU für die Dauer von 15 Minuten. Mit Hilfe einer speziellen Extraktionsmethode waren ca. 60% der applizierten Gesamtdosis im Perfusat nachweisbar, die systemische toxische Belastung war somit erheblich reduziert.
Nach Wiederherstellung der Blutzirkulation erholten sich die Tiere rasch und überlebten durchschnittlich 12 Wochen.
