Post culture behavior: genetic and epigenetic effects and related problems

Abstract

The occurrence of off-type plants propagated from tissue culture has caused concern among nurserymen (Anonymous, 1989; Evans et al., 1986; Knüttel, 1989; Read & Hosier, 1986; Stimart, 1986; Timmis & Ritchie, 1988; van Aartrijk & van der Linde, 1986). Typical off-type plants include dwarfs, color changes or mosaic patterns (e.g. chlorosis, loss of chimeras), growth habit changes (e.g. erectness, leaf shape, vigor), and changes in productivity (e.g. sterility, prolonged juvenility). Nevertheless, large numbers of plants are produced in vitro each year and typically variation among tissueculture-propagated plants from organized meristems is less than or equal to that observed from standard propagules (Ahmed et al., 1987; Ettinger & Preece, 1985; Hutchinson & Zimmerman, 1987; Jones 1988; Read & Hosier, 1986; Swartz & Lindstrom, 1986; Wu & Jampates, 1986). Recent advances in molecular genetics, as well as more detailed physiological and cytological study of the effect of tissue culture on genotypic stability and plant performance, has allowed a clearer understanding of the observed variation.
