Abstract

Photosensitive derivatives have been prepared corresponding to the biologically active analogs of the phenanthridine and acridine classes of DNA binding drugs. These were used as photoaffinity probes to study DNA binding, trypanocidal action, frameshift mutagenesis, mitochondrial mutagenesis, and DNA repair. In each instance there was good evidence that photoaffinity labeling provided covalent adducts corresponding to the noncovalent binding which led to the biological actions of the parent compounds, and the resulting covalent adduct formation resulted in significant enhancement of biological activity as confirmation. These studies illustrated the power of photoaffinity labeling for studying biological receptors of these important agents.
