High Speed Analytical and Preparative Separations of Biological Macromolecules

Abstract

A wide pore, 4000A, macroporous resin has been developed which is rigid and able to operate under HPLC conditions of flow rate and pressure. The poly(styrene/divinylbenzene) matrix can be used for reversed-phase separations and for both anion and cation exchange chromatography after surface coating/derivatization.

The open pore structure of this resin makes it suitable for the HPLC analysis of biological macromolecules due to the significant improvement in mass transfer. The rigidity and rapid equilibration enables small columns, 50 x 4.6mm, to be operated with steep gradients at high flow rates for both high resolution analytical separations and high load preparative fractionations.
