Development of DNA-assays for in Vitro differentiation and in Planta detection of Fusarium culmorum and Fusarium graminearum
Fusarium culmorum (W. G. Sm.) Sacc. (teleomorph unknown) and F. graminearum Schwabe (teleomorph Gibberella zeae (Schwein.) Petch) are causal agents of seedling rots, foot rots, leaf spots and head blights of small grain cereals (Cooke 1981). Their potential to produce mycotoxins harmful to the health of men and animals has drawn the attention to Fusarium seed contaminations. Simple and reliable diagnostic methods for both detection and quantification of Fusarium spp. are prerequisites for health risk evaluations. The diagnosis of Fusarium infections is based on visual assessment, microscopic or immunological techniques (ELISA). Simultaneous in planta quantification of distinct species is still not possible. Differentiation of Fusarium species relies on highly variable traits such as size and shape of conidia and conidiophores. Immuno- and DNA techniques are suitable to differentiate the forementioned species by species-specific antibodies or DNA probes and have the potential of simultaneous quantification. The objective of our work was the development of a reliable DNA- method for both in vitro differentiation and in planta detection of F. culmorum and F. graminearum.
