Abstract
We present results of five years of testing in-vitro cultures of over 30 apple and 20 stone fruit cultivars in a virus elimination programme. Our aim was to develop efficient large-scale screening designed for the special features of in-vitro plants to improve reliability in selection of truly negative in-vitro elite material following elimination treatment by thermotherapy and meristem preparation.
Commercial serodiagnostics were used in DAS or Two-Step ELISA. Screening for virus infection was generally carried out with 10–25 clones per cultivar and in accordance with the type of virus infections encountered, several cultivars were included in the investigations. Virus detection was undertaken at different growth phases during subculture, including short and long-term culture, a comparison of elimination treatments.
The following questions were considered:

Influence of subculture intervals

Can virus elimination measures just reduce virus titres, but fail to eliminate the virus. If so how long does it take for virus to reaccumulate to detectable levels?

How many tests are necessary to confirm initial test results? Are serological methods suitable for selecting truly virus-free progeny plants?
