DNA Microarray Technology Evaluation of Effect of Dietary Intake of Crude Rice Glycosphingolipids in Mice

Abstract

To determine the dietary effects of intake of crude rice glycosphingolipids (RG) extracted from rice bran, mice were fed a diet containing 0.5% RG for 2 weeks. Thereafter, we examined the gene expression patterns in the liver by using the DNA microarray technology. We observed that dietary intake of RG did not affect the expression of genes related to drug metabolism, showing that RG probably does not affect drug metabolism. The expression of genes related to glycolysis increased slightly, while that of genes involved in the TCA cycle decreased slightly. With regard to genes functioning in lipid metabolism, the expression of carnitine acetyltransferase (CRAT), which catalyzes acetylcarnitine production from acetyl CoA in a reversible reaction, markedly increased. The expression of the cytochrome P450 (Cyp) 7a1, which is involved in energy metabolism, was upregulated. These findings suggested that dietary RG promotes the metabolism of fatty acids and cholesterol metabolism by altering the expression of the CRAT and Cyp7a1 genes expression.
