Aminoacylase-Catalyzed Synthesis of N-Acyl Amino Acid from Fatty Acid or Its Ethyl Ester and Amino Acid

Abstract

There are many works on synthesis of valuable substances through condensation or transferase action by using hydrolytic enzymes, such as proteinases [1,2] or lipases [3]. However, few attempt has been reported for aminoacylase-catalyzed synthesis of N-acyl amino acid from amino acid and fatty acid or its ethyl ester [4]. Especially, fatty acid or its ester with the acyl-chain longer than 3 has not been used for the synthesis. In this study we tried to synthesize N-acyl amino acids with the acyl-chain of 2 to 7 from fatty acid or its ethyl ester and L-methionine or L-tryptophan by aminoacylase through condensation and transesterification. The products, N-acyl amino acids with longer acyl-chains, may be usable as safe biosurfactants.
