Analysis of sdi-1 Gene Functions in Human Malignant Gliomas

Abstract

A senescent cell-derived inhibitor of DNA synthetase-1 (sdi-1) occurs in human senescent cells. sdi-1 protein forms a complex with cyclin-dependent kinase 2 (cdk-2), and expression of sdi-1 is regulated by the p53 suppressor gene. We investigated the molecular changes of the sdi-1 gene in human gliomas and studied its effect on growth of glioma cells. DNA and mRNA were extracted from cultured human glioma cells and from clinical samples from patients with glioma and analyzed by Southern and northern blotting. The sdi-1 gene was transfected into cultured human glioma cells by a novel liposome method, and its growth inhibitory effect was investigated.
