Abstract
To examine the expression of vascular endothelial growth factor (VEGF), which may correlate with neovascularization in human brain tumors, the mRNA levels of the VEGF gene were determined by a Northern blot analysis in various human brain tumors, including gliomas, meningiomas, neurinomas, hemangioblastomas, hemangiopericytomas, and metastatic brain tumors. We obtained the VEGF mRNA expression index by dividing the VEGF mRNA levels by glyceraldehyde phosphate dehydrogenase (GAPDH) mRNA levels to compare the intensity of VEGF gene expression among tumors. In all brain tumors examined, the mRNA of VEGF was expressed at various levels. The mRNA levels of VEGF were significantly higher in gliomas and meningiomas. There was a significant difference between benign neuroepithelial tumors and malignant counterparts regarding the expression index of VEGF. However, the expression index showed similar values when anaplastic meningiomas were compared with meningotheliomatous meningiomas. The localization of VEGF protein was examined immunocytochemically. VEGF was specifically expressed on the endothelial cells in the tumor capillaries and also on the tumor cells surrounding them in various brain tumors. These findings suggest that VEGF is expressed in the wide range of brain tumors at various degrees and they may work on tumor angiogenesis in paracrine fashion to develop and maintain tumor vascularity. Furthermore, VEGF may work as an essential endothelial-specific mitogen in brain tumors.
