Zusammenfassung

Die methylotrophe Hefe Pichia pastoris (P. pastoris, Abb. 15–1) wurde in den letzten Jahren weltweit erfolgreich als System zur heterologen Genexpression etabliert und gehört mittlerweile neben Escherichia coli (Kap. 14), Saccharomyces cerevisiae, Hansenula polymorpha, dem Baculovirus-Expressionssystem (Kap. 14), Säugerzellsystemen (Kap. 12) und den zellfreien Systemen in vielen Laboren zum Standardrepertoire der Proteinproduktion (Cregg et al., 1993, Romanos, 1995, Higgins und Cregg, 1998, Gellissen, 2000, Cereghino und Cregg, 2000, Daly und Hearn, 2005, Schwarz et al., 2007). Neben einer Vielzahl löslicher Proteine wurden auch Membranproteine erfolgreich produziert und für biochemische sowie biophysikalische Untersuchungen isoliert (Reinhart und Krettler, 2006). Hierbei konnte sowohl für eine Reihe löslicher Proteine als auch für einige Membranproteine Material in ausreichender Menge und Qualität für Strukturanalysen produziert werden (Xu et al., 2002, Thoma et al., 2003, Binda et al., 2004). Auch die Isotopenmarkierung von rekombinanten Proteinen für NMR-Experimente wurde in P. pastoris bereits erfolgreich durchgeführt (Wood und Komives, 1999, Pickford und O’Leary, 2004).
