Zusammenfassung
Aus kleinsten bei Herzoperationen anfallenden Muskelstücken aus Vorhof und Ventrikel wurden menschliche Herzmuskelzellen isoliert. An diesen Zellen wurden mit Hilfe der patch-Elektrodentechnik Aktionspotentiale abgeleitet und in voltage-clamp Experimenten die diesen Erregungsprozessen zugrundeliegenden Ionenströme analysiert. Während für die Depolarisations-phase von Vorhof und Ventrikel ein schneller Natriumstrom in Verbindung mit einem Kalziumstrom mit langsamer Kinetik verantwortlich ist, zeigte sich, daß in der Repolarisationsphase ein deutlicher Unterschied zwischen den beiden Gewebetypen besteht. Hier spielt im Vorhof ein transienter Kaliumstrom, im Ventrikel jedoch der für diesen Herzbereich typische “delayed rectifier”-Kaliumstrom in Verbindung mit dem “inward rectifier”-Kaliumstrom die entscheidende Rolle. Die vorliegenden Resultate zeigen, daß an der menschlichen Herzmuskelzelle physiologische und pharmakologische Untersuchungen mit den modernsten biophysikalischen Methoden möglich sind, welche dieses Präparat zu einer wichtigen Alternative zu Versuchen mit Gewebematerial aus Versuchstieren machen.
