Summary
Ribavirin or Virazole (1-β-D-ribofuranosyl-1,2,4-triazole-3-carboxamide) is a broad-spectrum antiviral agent whose molecular mode of action remains controversial. Results of earlier experiments indicated that ribavirin possesses a significant direct suppressive effect on the vesicular stomatitis virus (Indiana) (VSI) viral polymerase and that this inhibitory activity appears to block initiation of transcription. This direct effect can be mediated by the mono-and diphosphorylated forms of the drug as well as the triphosphorylated form. To confirm our previous results we examined the effects of analogs which stabilize as diphosphates on this VSI virus in vitro transcription reaction; all exhibited inhibitory activity to varying degrees. The results of these studies are discussed in relation to inhibition of primary transcription and its effect on decreased viral progeny production.
